A versatile plasmid architecture for mammalian synthetic biology (VAMSyB)

چکیده
Current molecular cloning strategies generally lack inter-compatibility, are not strictly modular, or applicable to engineer multi-gene expression vectors for transient and stable integration. A standardized platform would advance research, example, by promoting exchange of between groups. Here, we present a versatile plasmid architecture mammalian synthetic biology, which designate VAMSyB, consisting three-tier vector family. Tier-1 is designed easy engineering fusion constructs, as well swapping genes modules tune the functionality vector. Tier-2 expression, constructed directly transferring engineered cassettes from tier-1 vectors. Tier-3 enables integration into host cell through viral transduction, transposons, homology-directed recombination via CRISPR. This VAMSyB expected have broad applicability in field biology. The collection plasmids will be available Addgene.

متن کاملVersatile Design and Ethical Architecture
Background: Following the spread of dissatisfaction with modern architecture, the machine attitude towards man, and the disregard for human values and human existential dignity, the global movement in support of independence and participation was formed in response to demographic and social realities. Recognized in 1988 as universal design, the movement allowed more people of all levels of ab...
متن کاملSystems approaches for synthetic biology: a pathway toward mammalian design
We review methods of understanding cellular interactions through computation in order to guide the synthetic design of mammalian cells for translational applications, such as regenerative medicine and cancer therapies. In doing so, we argue that the challenges of engineering mammalian cells provide a prime opportunity to leverage advances in computational systems biology. We support this claim ...

متن کاملA versatile plasmid system for reconstitution and analysis of mammalian ubiquitination cascades in yeast
Ubiquitination is a posttranslational protein modification that regulates most aspects of cellular life. The sheer number of ubiquitination enzymes that are present in a mammalian cell, over 700 in total, has thus far hampered the analysis of distinct protein ubiquitination cascades in a cellular context. To overcome this complexity we have developed a versatile vector system that allows the re...
